Yes. There is a dedicated boarding point at JR Kyoto Station Hachijo Exit sightseeing bus parking lot at 9:50 AM, so you do not need to travel to Osaka first.

Are the boat tour and cycling included in the price?
No — these are optional activities payable on the day at Ine Funaya and Amanohashidate respectively. The Amanohashidate Onsen is also at your own expense.
